De novo biosynthesis of ?-cadinol in engineered Escherichia coli

نویسندگان

چکیده

Abstract ?-Cadinol is a sesquiterpene that widely used in perfume, fine chemicals and medicines industry. In this study, we established biosynthetic pathway for the first time engineered Escherichia coli production of ?-cadinol from simple carbon sources. Subsequently, further improved to 35.9 ± 4.3 mg/L by optimizing overproduction rate-limiting enzyme IdI. Finally, titer was increased 133.5 11.2 with two-phase organic overlay-culture medium system. This study shows an efficient method biosynthesis E. heterologous hybrid MVA pathway. Graphical

De novo Biosynthesis of Biodiesel by Escherichia coli in Optimized Fed-Batch Cultivation

Biodiesel is a renewable alternative to petroleum diesel fuel that can contribute to carbon dioxide emission reduction and energy supply. Biodiesel is composed of fatty acid alkyl esters, including fatty acid methyl esters (FAMEs) and fatty acid ethyl esters (FAEEs), and is currently produced through the transesterification reaction of methanol (or ethanol) and triacylglycerols (TAGs). TAGs are...

Modeling allosteric regulation of de novo pyrimidine biosynthesis in Escherichia coli.

With the emergence of multifaceted bioinformatics-derived data, it is becoming possible to merge biochemical and physiological information to develop a new level of understanding of the metabolic complexity of the cell. The biosynthetic pathway of de novo pyrimidine nucleotide metabolism is an essential capability of all free-living cells, and it occupies a pivotal position relative to metaboli...

Studies on the de novo Biosynthesis of NAD in Escherichia coli

Quinolinic acid (pyridine 2,3-dicarboxylic acid) which is an immediate precursor of the pyridine nucleotides, is synthesised from L-aspartate and dihydroxyacetone phosphate in Escherichia coli. Extracts from certain nadB mutants complement the extracts prepared from all nadA mutants for the enzymic synthesis of quinolinate. Using the complementation assay, the quinolinate synthetase B protein h...

Artificial de novo biosynthesis of hydroxystyrene derivatives in a tyrosine overproducing Escherichia coli strain

BACKGROUND Styrene and its derivatives as monomers and petroleum-based feedstocks are valuable as raw materials in industrial processes. The chemical reaction for styrene production uses harsh reaction conditions such as high temperatures or pressures, or requires base catalysis with microwave heating. On the other hand, production of styrene and its derivatives in Escherichia coli is an enviro...

Inhibition of de novo purine biosynthesis and interconversion by 6-methylpurine in Escherichia coli.

The inhibition of Escherichia coli strain B and strain W-11 by 6-methylpurine depended on the formation of 6-methylpurine ribonucleotide by the action of adenine phosphoribosyltransferase (AMP: pyrophosphate phosphoribosyltransferase, EC 2.4.2.7). 6-Methylpurine ribonucleotide inhibited the de novo synthesis of purines, presumably via pseudofeedback inhibition of phosphoribosylpyrophosphate ami...
